Asaf is right, E=mc2 is the rest energy of mass m. This equation has nothing to do with the determining the speed of light c. Speed of light is given by the current properties of vacuum (or space-time, if you wish). And because vacuum (space-time), as we know it, was created before or around the Plank time, the answer why c= 3x10^8 should be in the conditions and processes in the Universe back then. People are still working on that.
